Detection of Protein Aggregation using Fluorescence Correlation Spectroscopy

Protein aggregation is a hallmark of neurodegenerative disorders such as amyotrophic lateral sclerosis (ALS), Alzheimer's disease (AD), Parkinson's (PD), Huntington's (HD), and so on. To detect analyze soluble or diffuse protein oligomers aggregates, fluorescence correlation spectroscopy (FCS), which can the diffusion speed brightness single particle with molecule sensitivity, has been used. However, proper procedure know-how for detection have not widely shared. Here, we show standard FCS measurement properties aggregation-prone proteins in cell lysate live cells: ALS-associated 25 kDa carboxyl-terminal fragment TAR DNA/RNA-binding 43 (TDP25) superoxide dismutase 1 (SOD1). The representative results that part aggregates green fluorescent (GFP)-tagged TDP25 was slightly included fraction murine neuroblastoma Neuro2a lysate. Moreover, GFP-tagged SOD1 carrying mutation shows slower cells. Accordingly, here introduce to via its property using FCS.
